Preface

This Book is published as a mid-term step (i.e., middle of every 3-year period) by the Allergy and Immunotoxicology Scientific Committee (AISC) of the International Congress of Occupational Health (ICOH). The comprehensive meeting of ICOH is held every 3 years. Recently, it was held in Seoul, South Korea, 31 May–5 June 2015, when Prof. Mario Di Gioacchino, of Chieti, Italy, served as the chairperson. The next meeting will be held in Dublin, Ireland, in 2018. In conjunction with the opening of the special, general, oral, and poster sessions as well as joint sessions with other scientific committees (SCs), several SCs are staging mid-term activities independently. Our SC/AISC has also been holding several special midterm symposiums in Italy, China, and Japan (in Kumamoto and Kyoto) and calls each such symposium an International Symposium on Occupational and Environmental Allergy and Immune Diseases (ISOEAID). As the current chairperson of this SC, Prof. Otsuki was the local organizer of ISOEAID 2010 held at Kyoto. It was April and we were surrounded by beautiful cherry blossom trees in full bloom. During the symposium, we presented and discussed many issues regarding the occupational and environmental allergy and immunotoxicology field and enjoyed the lectures on air pollution such as the effects of PM2.5 on immune organs, immunological aspects of sick-building syndrome, sensitizers in GHS (The Globally Harmonized System of Classifi cation and Labelling of Chemicals), and childhood and immunology.
The first honorary chairperson in our SCs was Prof. Toshio Matsushita, Kagoshima, Japan. Thereafter, Prof. Poalo Boscolo, Chieti, Italy, and Prof. Kanehisa Morimoto, Osaka, Japan, inherited the chairperson’s role. All three editors of this eBook have been in that role in our SC. Prof. Di Gioacchino served as Chair 2009–2014. I have been the Chair since 2015, with Prof. Petrarca as the secretary in the SCs. Subsequently we decided to publish this eBook instead of ISOEAID at this mid-term point based on the presentation in the ICOH meeting in Seoul, South Korea.
The AISC has been focusing on investigation and discussion of the pathophysiological mechanisms involved with the allergic and immunotoxicological effects of environmental as well as occupational substances. Particularly, occupational allergies such as dermatitis and asthma were practically classical occupational diseases. The basic strategy is to avoid exposure, whereas there have been numerous newly developed substances that are being introduced in occupational situations. Thus, establishing methods of prediction using in vitro and in silico procedures is required. In addition to these investigations, epidemiological studies are also important, as is establishing a system to quickly identify allergic diseases caused by newer substances, and it is valuable, as well, to prevent workers from contracting these diseases. In addition, with allergic diseases the immunotoxicological effects of various substances in the environment as well as occupational settings are also important. Major members of our SC have been investigating the immunotoxicological effects of fibrous and particulate matters such as silica, asbestos, and recently nanomaterials including nanoparticles, nanotubes, and nanosheets.

Preface

ADP-ribosylating toxins have been the focus of intensive research for more than 30 years. Researchers from diverse fields of science have taken an interest in these bacterial toxins; they are studied, for example, by microbiologists, biochemists, cell biologists, and pharmacologists. There are two principal reasons for the broad and still growing interest in ADPribosylating toxins. First, insights into the structure and functions of the toxins might be the key to prevention and treatment of diseases caused by the toxin-producing infectious microorganisms. Second, the ADP-ribosylating toxins provide potent and often unique pharmacological tools for the study of the physiological functions of their target proteins. The latter is especially the case with cholera and pertussis toxins, which both modify the IX-subunits of heterotrimeric G-proteins involved in signal transduction pathways. These toxins have proved invaluable in extending our basic understanding of the regulation of hormone-controlled signal transduction.
This volume provides a review and an update of recent studies on the basic properties of bacterial ADP-ribosylating tbxins and/or exoenzymes. Our current knowledge of the cellular entry mechanisms of ADP-ribosylating toxins is reviewed by MADSHUS and STENMARK. WILSON and COLLIER then deal with recent insights into the enzyme mechanism and active site structure of diphtheria toxin and Pseudomonas aeruginosa exotoxin A, which modify elongation factor 2. Toxins which ADP-ribosylate heterotrimeric G-proteins involved in transmembrane signal transduction are the subject of the next two chapters. SERVEN11, Moss and VAUGHAN describe the regulation of adenylate cyclase by cholera toxin and the enhancement of the cholera toxin-induced ADP-ribosylation by guanine nucleotide-binding proteins, the so-called ADP-ribosylation factors, and the review of pertussis toxin by GIERSCHIK lays emphasis on toxin structure-function relationship, toxin-Gprotein interactions and the functional consequences.of G-protein ADP-ribosylation. A chapter by AKTORIES, WILLE, and JUST discusses recently described actin-ADP-ribosylating toxins such as Clostridium botulinum C2 toxin, C. perfringens iota toxin, and other iota-like toxins. Together with the mycotoxins cytochalasin and phalloidin, these ADP-ribosylating toxins appear to be important new tools with which to study the role of actin in various physiological processes. AKTORIES, MOHR, and KOCH deal with clostridial exoenzymes which modify small GTP-binding proteins. C. botulinum C3 ADP-ribosyltransferase, C. limosum exoenzyme, and their eukaryotic protein substrates Rho and Rac are discussed in detail. Last but not least, COBURN briefly reviews P. aeruginosa exoenzyme S. Much less is known about this ADP-ribosyltransferase and its eukaryotic substrates. Recent studies indicate that, again, small GTP-binding proteins (Ras proteins) are the preferred substrates of the enzyme.
This book offers an exciting and comprehensive account of recent developments in the field of ADP-ribosylating toxins and should inspire both further research in this area and the use of these toxins as tools in biological science.

We live and work in the age of immunotherapy. The modality is now firmly affixed to the triad of chemotherapy, radiation therapy, and surgery. This book captures the translation of immunology into therapies. The migration of bench research to bedside experimentation has been largely driven by academia and amplified by industry; however, in the current age, there is equipoise between the not‐ for‐profit and for‐profit enterprises regarding advancements in the human applications of immunotherapies. The breadth of treatments reflects the complexity of the immune system itself. The coordinated response of the multiple components of an endogenous immune response has generated a portfolio of immunotherapy options that are reflected in the names of the chapters. Not all chapters, though, are created equally. Some immunotherapies are just beginning their human experimentation and some are seasoned and perhaps even seen as out of vogue. Nevertheless, as a whole, these components of immune‐based therapies provide patients with therapeutic optimism and some with therapeutic impact. This book is a sum of its chapters and thus individual immunotherapies. What is not yet evident is how combinations of immune‐based therapies can be harnessed. This is undoubtedly needed in order to secure long‐term and complete treatment for the majority of malignancies, especially arising as solid tumors. The coordinated response of the endogenous immune system will be mirrored by the corradiated application of immunotherapies. However, that will be the topic of a future book. What is present and is remarkable, is that monotherapies based on harnessing the immune response have resulted in Lazarus‐type moments, are used to prevent cancer, and have provided responses in tumors that were previously considered untreatable. Immunotherapy as presently wielded is a relatively blunt tool. Yet the immune system is built on precision. Academics and industry investigators are only beginning to understand how to sharpen the therapeutic edge of an applied immune response. The proving ground is the human experience as preclinical models by and large do not yield sufficient information regarding efficacy and toxicity. Thus, immunotherapy practitioners and patients alike are risk takers. Together, they will advance the clinical application of the immune system so that its complexity can be harnessed as an instrument to treat cancer on an individualized basis.
